Background

KRAS is a small GTPase protein located at the inner cell membrane, functioning as a molecular switch that regulates cell proliferation and survival signals. The Q61H mutation resides in the Switch II region, impairing GTPase activity and locking the protein in a constitutively GTP-bound "active" state, leading to aberrant activation of MAPK/PI3K pathways. This mutation is commonly found in pancreatic, colorectal, and non-small cell lung cancers. Compared to KRAS G12 mutations, Q61H exhibits resistance to SHP2 inhibitors and is associated with worse prognosis. Clinical detection of this mutation provides crucial guidance for targeted therapy selection and prognostic evaluation.

Assay protocol

Principle: The GTPase Glo™ Assay assesses the activities of KRAS by detecting the amount of GTP remaining after GTP hydrolysis in a KRAS reaction.

Materials

1.KRAS(Q61H) His Tag Protein, Human

2.GTPase Glo™ Assay (Promega, Catalog # V7681T)

3.Solid white multi-well plate (384-well plate) (Corning, Catalog #3572)

4.Plate Reader (PerkinElmer)

Produce

1.Prepare a 2X GTP solution containing 10 µM GTP and 2mM DTT in GTPase/GAP Buffer.

2.Dilute the KRAS to 100 µg/mL, 80 µg/mL and 60 µg/mL in GTPase/GAP Buffer and dispense 5 µL into each well of a 384 well plate.

3.Initiate the reaction by adding 5 µL of the 2X GTP solution prepared in Step 1 to each well. Include a 2X GTP solution with 5 µL GTPase/GAP Buffer as Blank. The reaction volume is 10 µL.

4.Incubate the reaction at room temperature (22-25℃) for 30 minutes.

5.Gently mix the thawed GTPase Glo™ Reagent, 500X, by inversion; do not vortex. Prepare the required volume of reconstituted GTPase Glo™ Reagent by increasing or decreasing the component volumes provided below.

6.Add 10 µL of reconstituted GTPase-Glo™ Reagent to the completed reaction, mix briefly and incubate with shaking for 30 minutes at room temperature (22–25℃).

7.Add 20 µL of Detection Reagent and incubate the plate for 5-10 minutes at room temperature (22–25℃).

8.Read at emission wavelengths of 555 nm (luminescence), respectively in endpoint mode.

9.Calculate specific activity.

Excellent Time Keeper // Terrible Wristband !
The watch itself is fantastic. When I first got it, and set it up, I set it to the time on my computer, which is as accurate as you can get. In the beginning I checked the time accuracy daily. After a few weeks my time checks changed to once, or twice a week. I never had to change the watch's time. The seconds would vary a little, in comparison to the computer clock. But, it never got more than 30 seconds either side of the computer clock. However, the watchband is a big time NO-GO! The buckle, where you connect it on your wrist, can be frustrating most of the time when you are trying to put it on. Once you've got it on, the strap that goes beyond the buckle is held down by a sliding keeper, that slides in and out depending on what you're doing. Terrible design! I wanted to buy a "Twist-a-Flex" wristband, but couldn't find one that matched the unusual design where the straps attach to the watch body. I bought the watch 4 months ago. Today, one of the 4 pins, that connect the straps to the watch body, fell out somewhere, making the watch impossible to wear. Excellent price. Excellent timepiece. Worst design for a wristband that I've ever seen.

Excellent for price, you won't be sorry
If you are looking for a watch with a lot of features that doesn't cost a lot, this is your watch. I tear up watches fairly easily in my line of work and after I killed my Seiko I decided I would hunt out a cheap watch. There aren't a lot of watches at this price range that include a stopwatch, date (with day of the week), AND has a light that is bright enough to actually read the time in the dark. This one does. The watch not dainty; it is big and bulky, bigger than I thought it would be, but not unwieldy and not super heavy. Expect it to be big. This also helps with readability (My son can read the time on this watch from the other side of the couch). The band feels rugged, made of a pliable and soft- feeling rubber and has a two- pronged belt- like closure (think 'biker belt') that keeps it secure on the wrist. Additionally, the holes for the band goes the entire length of the band so it is adjustable for really small wrists up to about 9.25" circumference. Others have complained about how easily the face scratches, but I suspect they've failed to remove the protective coating on the lens. It's a protective film that you have to get your fingernail under to pull off. Once that's off, the lens is crystal clear and fairly tough. I've had mine for a week now and used it under some fairly harsh conditions and the lens is still clear and scratch-free. My only complaint seems petty for a watch at this price point, but the strap-end hold down 'belt'--- the loop that holds down the extra length of the strap when it's buckled--- moves easily and so in use it will occasionally come off and you have to retuck the end back into it. But seriously, this is really minor. If you need an inexpensive digital that's easy to read in both daylight and darkness, ya don't mind a large and bulky, rugged- looking watch, and don't want to spend an arm and a leg, get this one. You likely won't be sorry.
